pro's CI - 10:10pm

The furnace guy was going to come over tonight to see if it really was 62 degrees in here as I said, so I cleaned up the place. He didn't show up, but now I have a neat apartment. I even worked on my pile for 15 minutes or so.

I should get ready for bed now - brush my teeth, etc.

10:15 CI. Lark

Man, it's cold here this morning too. The forecast is very nice for the weekend, but I don't trust myself that much to put off my work till then. You know how "things come up". My workshop is heating slowly, and when I log off here, I'm going to start cleaning there till it's nice enough to sit still and work. I hope you get heat by the time you make it home tonight, Pro!

Sitting in the doctors office waiting for dh to have everyones favorite medical test.

The first order of business today is to get all my disorganized lists back in GTD shape. I got my email organized again before the holiday and I have done away with the paper inbox altogether. But my todo list and therefore my mind is in a mess.

When I have a doable GTD action list it's much easier to, well, Get Things Done. When I don't have that, I tend to get off track. Its too easy to drift off after finishing one task because its not clear in my mind what I want to do next. And my mind just wanders off...

So:
- Make a realistic to do list
- Organize to do lists - 2 hrs
- doc and medical stuff - 2 hrs
- food shopping - 1 hour
- fun shopping - 1 hour
- straighten house - 1 hour
- set up sewing machine - .5
- 1 item of mending - .5

- dinner 1 hour
- free time 2 hour

Didn't get the mending done, but I did go to the craft store and get buttons for one of my favorite suits that is missing a button.  Got one out of two piles of junk cleaned up, and I'm done for the day. 

A much better day than yesterday, productivity-wise!

On time to work (well, a few minutes late but I'm the first one here)
I'm going to start by finishing my task from yesterday...mapping TFs to geneIDs. When I'm done with that, I'll check in and then make my to-do list for the rest of the day.

1)Get sequences corresponding to annotated genes (45 minutes)
2)Start THEME running on these sequences (about 45 minutes to start the jobs)
3)Figure out how to use new interactome with flow algorithm, run test job (1.5 hrs)
4)get geneIDs of coregulators (1 hr)
5)Analyze THEME results (1 hr)
6)Start NDSEG essay (2 hr)
7)Go running (1 hr)
8)Go to Harambee (3 hrs)
and I'll figure out what to do with my evening when I get home

OK, a bit out of order, I got the geneIDs of coregulators first.
Now I'm going to go get a soda (I need my caffeine jolt), then actual get the sequences like I'm supposed to.

OK, getting sequences is actually proving to be quite difficult. I think I've finally figured out how to do it, but the method is a bit convoluded :-P
Method:
1)Map geneIDs to HGNC accessions (15 min)
2)Map HGNC accessions to RefSeq IDs (15 min)
3)Use RefSeq IDs to get locations in genome (15 min)
4)Use genome locations to actually get sequences (15 min)
5)Take notes on process (20 min)
Whew! That's a mess. I'll be sure to automate it/take lots of notes, so I won't have to go through this pain again :)
This is deadly dull, so I'll  check in frequently so I stay on track

Finally got the sequences, tedium it over!
Now I can start to analyze then.
Will take a short break (<10 min).
Then, I'll start THEME program running on then; should take  45 min-ish to set up/start
Will check back when program is started

EDIT 2:25: a little processing needs to finisj before I can actually launch the program, but effort on my part is done.

I'll spend 1 hous figuring out how the flow algorithm works with the new interactome.
Then, I'll move onto working on my NDSEG essay (since that's due the 7th, argh!)
At 5, I'll stop work, and go running and then on to Harambee.
When I get home (around 8:30 or 9), I'll analyze the THEME results. If I'm not too tired, I'll work more on NDSEG.

EDIT: 3:00 Got a bit stuck on working with the new interactome, but I've decided that my main task is to modify my code to use Xgraphs. I'll start with a small (4-5) page portion of code, working from a printout as much as possible so I stop being distracted by the internet.
I'm losing focus, so after about 30 minutes I'll go out running, then come back to work. Check in again after the run.

Sorry for the huge number of check-ins!

I went running...it went a bit long, but felt good.
Now I'm going to spend 45 minutes working on NDSEG.
Probably won't get anything finished, but it need to get over the notes->actual writing hurdle, so I'll write with a timer. 
I'll stop promptly at 5:45, so I can be on time to Harambee

Went to Harambee meeting, then ended up going out a drink with friends.
I'm feeling pretty exhausted, but I need to get stuff done anyway :(
However, I'm going to go to bed promptly at 12.

30min checked over NDSEG app (I'm too tired to work on essay, but I think I can proofread)
40min going through finances
30min organizing papers
10 min reading Bible
Read, set, go!
EDIT: 11 I'm completely exhausted, am going to go to bed and get up early.
